Hexamethylphosphoramide undergoes cytochrome P-450 -mediated N-demethylation to Formaldehyde. Hexamethylphosphoramide induces DNA-protein crosslinks . Hexamethylphosphoramide has been linked to nasal tumors (squamous cell carcinoma, adenoid squamous cell carcinoma), squamous metaplasia, rhinitis, tracheitis, and reversible and irreversible infertility.
