E12CA1A3 is an ionizable cationic lipid (pKa = 6.4) used to generate lipid nanoparticles (LNPs) for mRNA delivery in vitro and in vivo. LNPs formulated with E12CA1A3 and an mRNA reporter successfully induce luciferase expression in both mouse bone marrow-derived dendritic cells (BMDCs) and live mice. Additionally, E12CA1A3-based LNPs exhibit faster clearance from the liver compared to DLin-MC3-DMA-based LNPs in mice.
