Lipid HTO12 is an ionizable lipid (pKa = 6.432) used in lipid nanoparticles (LNPs) for mRNA delivery. LNPs with HTO12 and adenine base editor mRNA targeting Pcsk9 reduce hepatic Pcsk9 levels in mice, requiring a lower lipid-to-mRNA ratio than SM-102-based LNPs.
