Lipid ND-O1 is an ether-containing ionizable cationic lipid (pKa = 6.6) that has been used in the generation of lipid nanoparticles (LNPs) for the delivery of siRNA in vitro and in vivo. LNPs containing ND-O1 and encapsulating Cy5-labeled siRNA selectively accumulate in the liver over the spleen, heart, kidney, and lung in mice.
