Dehydroergosterol (DHE), a phytosterol derived from ergostane, contains multiple double bonds at positions 5,6-, 7,8-, 9,11-, and 22,23-, along with a 3beta-hydroxy group. It is a highly rigid C-24 methyl derivative with one double bond in the tail region and two double bonds in the body region. Being a C-24 alkyl derivative, its tail and body variances influence the size, mRNA encapsulation efficiency (EE), and transfection efficiency of lipid nanoparticles (LNPs).
