LD003 (solution in ethanol)

Overview

LD003 is a lipid prodrug derived from the synthetic glucocorticoid dexamethasone, used in the formulation of lipid nanoparticles (LNPs) for in vivo siRNA and mRNA delivery. LNPs containing LD003 and encapsulating cytosine-guanine-containing oligodeoxynucleotide (CpG) reduce blood levels of IL-10, CXCL1, TNF-α, IL-1β, and IL-6 in mice. Additionally, LNPs with LD003 and siRNA targeting mouse coagulation Factor VII (FVII) decrease immune cell infiltration at the injection site and lower hepatic FVII levels in mice.
