8-Shogaol is a phenolic ketone and one of the major pungent compounds in ginger (Zingiber officinale), characterized by a 14-carbon alkyl side chain. It is a potent TRPV1 agonist that alleviates acute and inflammatory pain by activating and subsequently desensitizing the TRPV1 channel, a mechanism independent of opioid receptor function. 8-Shogaol effectively reverses morphine analgesic tolerance and reduces nociceptive behaviors elicited by chemical stimuli, noxious heat, and inflammation. Key binding sites for its interaction with TRPV1 include Leu515, Leu670, Ile573, Phe587, Tyr511, and Phe591. The compound also exhibits anti-inflammatory activity through TAK1 inhibition (IC₅₀ = 5 μM), COX-2 inhibition (IC₅₀ = 17.5 μM), and NF-κB pathway suppression. Additionally, it shows antiplatelet aggregation, anti-rheumatoid arthritis, and potential anticancer properties.
