Ganoderic acid A

Overview

Ganoderic acid A is a highly oxidized lanostane-type triterpenoid isolated from Ganoderma lucidum (Reishi mushroom), one of the earliest and most abundant ganoderic acids discovered. It exhibits broad pharmacological activities including anticancer, anti-inflammatory, hepatoprotective, lipid-lowering, neuroprotective, and anti-aging effects. Ganoderic acid A targets glucose transporters GLUT1 and GLUT3 to inhibit cancer cell glucose uptake, and modulates multiple signaling pathways including NF-κB, JAK/STAT, PI3K/AKT, and MAPK to induce apoptosis and cell cycle arrest. It also functions as a senotherapeutic agent that reduces cellular senescence markers, extends lifespan in C. elegans by 26%, and improves healthspan in aging mouse models. Recent studies have identified it as a p53-MDM2 pathway regulator, with its amide derivatives showing enhanced anticancer activity through MDM2 binding (KD = 1.68 μM).
